It's kind of an anti-joke, the lead up infers that you would fill the hole with something sappy like love, but people try and do it with material things, religion, casual sex, etc.

And then it just says something silly, which could be construed as deep... I suppose.

Not too much itself, but it is a popular story religious people like to tell children. That everyone has a hole in their heart that makes them feel empty and hollow; some people try to fill the emptiness with money, fame or drugs but the only thing that works is to "let Jesus in your heart". I was told this many times when I was forced to attend church. It was a surprise to me, I didn't know I had this hole until they told me and when they told me I started to feel it... It was only after I stopped trying to awkwardly force religion in it that I realized I never had one in the first place. Fucking liars.

It's basically just existentialism. Instead of searching for meaning, be okay that there is none and find your own way to enjoy that. I'm not sure whether he was going for the humor of exactly what you said and only did the existentialism angle by accident, or if he was trying to present an existentialist position and tossed some humor in on top.

My favorite theoretical religion is "democratic religion". When someone is elected to office, the voters bestow powers to them. Magical powers. And control over increasing numbers of mindless flunkies the higher the office. But the higher the office, the more insane it makes them.

So like, a mayor can magically fix a traffic ticket, and just has a few mindless secretaries working for them, and is eccentric; but the president can use their powers to twist and reshape reality itself, ruling over vast armies of Orcs, but they are so bonkers it always comes out weird and distorted.

So what a democratic religious person does is make an effort to avoid elected officials and their mindless flunkies. And when they see them or meet them to remember that they might turn you into a newt or something, but they cannot help themselves because they are nuts, so be polite.
