Simple explanation: Japan.

Longer version: Essentially, there's a challenge (in this case, a race), and the losers have to participate in the punishment game. Except for the 24h Tag, there is a theme to it (Flight attendants, police officers, nurses, etc.) and has many traps associated with it. The 24h Tag is different from the rest because, as I recall, it was the first one ever so it didn't have most of the mainstays of the normal series. In the Tag game, if they get caught, they get punished; in every other 24h Punishment game, if they laugh, they are punished. In the No Laughing Games, the participants try to make the others laugh as well since they're assholes. It's pretty much co-workers being dicks to each other with the dial turned up to 11.

Oh man, they are the funniest nurses ever. On one of them they have to get into an MRI and only once almost fully inside do they notice that there's an older woman's face in the MRI machine that ends up right above them, and then the table they're on slowly raises until she can kiss them. Every one of them failed that time.

Kinda but not really. Of course they understand that they need to put on a show for the camera, they are entertainers after all. But things like the punishments (like the hotel one where they got blowdarted in the ass) all of those reactions were real and they knew nothing of the events which made them laugh beforehand.

Some of the batsu games though, like the one where Hamada goes to Russia, a lot of that is scripted. But once again, all the reactions are real and he has no idea going into it what will happen.

One of the main guys from Gaki no Tsukai. Its a show in Japan thats been going on for like 15 years or something. The shows basic premise is funny shit like skits (as seen in the gif) and batsu games (punishment games) where you get punished for laughing or w/e. Blowdarts in the ass, smacked by a wooden rod, and etc are all popular punishments.

Btw the show is fucking hilarious, easily one of the best shows in the world.

He and his partner Hamada (the one on the other side of the glass) are credited with revolutionizing manzai, a type of comedy routine performed in Japan by a duo. They're in pretty much in everything, tv, music, movies you name it. They are household names in Japan when it comes to comedy very respected since they've been in the biz for so long. Hard to compare to American comedians because none have careers on top as long as these guys.
